The Human LIF (Leukemia inhibitory factor) QuickTest ELISA Kit measures LIF in Human Serum, Plasma, Cell Culture Supernatant, Cell Lysate or Tissue Lysate. This kit was based on sandwich ELISA method. The experiment lasted 120 minutes. Capture antibody was conjugated to an affinity tag that was recognized by a specific antibody coated on the QuickTest plate. Add the Cap/Det Ab working solution into each well, then add the standards and pilot samples into individual wells. If the sample contains LIF, a capture antibody-LIF-biotin-detection antibody complex was formed. After incubation, unbound conjugates were removed by wash buffer. HRP-Streptavidin was added. After washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of LIF in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.

Won't last forever, but my dog loves it

My dog, a 90 lb black lab, LOVES these toys. He does prefer the raccoon to the giraffe, but he likes them both a lot. I originally got the raccoon on clearance at Target when looking for a long toy he could play with while he was in a cone recovering from surgery (he likes to be able to hold toys between his front paws and mouth/chew on them, so none of our regular squeaky footballs were long enough for that). He loved it so much I had to find more, so I found this giraffe from the same line of toys. He's a destroyer and he has been able to put a few holes in these toys if I'm not watching him, but they're pretty easy to sew if they need patching. My mother-in-law's dog came over and made a hole so big that the squeakers and lining all came out, but I sewed it back up and it's almost as good as new. They definitely won't last forever, but they have lasted longer than other soft toys we've had in the past.
